Abstract
This study was included the isolation of Bacillus subtilis bacterium from soil samples and diagnosing by traditional methods and VITEK-2 system, to improve its productivity by manipulating the components of the production medium and environmental conditions, were studied using several natural carbon sources.
Followed by genetic improvement of the selected isolate Bacillus subtilis EN3 by applying protoplast fusion technique. On the other hand, the mutation by ultraviolet light was carried out to the fusant isolate B.subtilis EN3A1-P19 to enhance the glutamic acid production. And this isolate was the optimal one compared with the other fusant and UV light mutant isolates. Glutamic acid was extracted from production medium, crystalized, purified and characterized using HPLC and FTIR.
