Abstract

An isolate of local bacteria which efficiently distinguished in alkaline protease production was obtained. This enzyme  was used in some of application fields which requires to be  active in the alkaline conditions. This isolate was identified  a Bacillus cereus  MH12 based on morphological, cultural characteristics, Vitek2 system and  genetic analysis. To increase alkaline protease productivity  from this locally isolate, the optimum  condition for enzyme production  has been studied by submerged state fermentation, then the enzyme was partially purified by precipitation with 30-70 % of saturation of ammonium sulfate followed by dialysis and ion-exchange chromatography, finally the enzyme extract was subjected to gel filtration.

The characteristics of the purified enzyme was studied in order to identify it’s suitability when it use in  some industrial applications. These characteristics were included optimum pH and temperature for activity and stability, enzyme specificity toward some of substrates  and effect of some ionic  metals  on enzyme activity. The efficiency of Alkaline protease produced from local isolate  Bacillus cereus  MH12  were studied  in removing of proteins spots  from cotton cloth  and  compatibility of enzyme used with some of  commercial detergent powder in local markets . The enzyme  also used with high efficiency  as alternative to chemicals such as sodium sulfide 2% and calcium  oxide 10%  to remove hair from cow skin when  treat skin  with   100 % enzyme   ( 612.2 U/ ml ) for 24 h at 50 0 C.
